چکیده
Cho et al. (2021) and Villa demonstrate that mTORC1 stimulates m6A mRNA methylation via WTAP expression SAM synthesis. Increased in turn promotes cell growth by enhancing degradation or translation.

m6A mRNA Methylation: A New Circadian Pacesetter
The purpose of m(6)A methylation of RNA, first observed in the 1970s, has been a longstanding mystery. Fustin et al. now show that it regulates RNA processing and determines the period and oscillatory stability of the mammalian circadian clockwork.
متن کاملm6A-Driver: Identifying Context-Specific mRNA m6A Methylation-Driven Gene Interaction Networks
As the most prevalent mammalian mRNA epigenetic modification, N6-methyladenosine (m6A) has been shown to possess important post-transcriptional regulatory functions. However, the regulatory mechanisms and functional circuits of m6A are still largely elusive. To help unveil the regulatory circuitry mediated by mRNA m6A methylation, we develop here m6A-Driver, an algorithm for predicting m6A-driv...
متن کاملPerturbation of m6A writers reveals two distinct classes of mRNA methylation at internal and 5' sites.
N6-methyladenosine (m6A) is a common modification of mRNA with potential roles in fine-tuning the RNA life cycle. Here, we identify a dense network of proteins interacting with METTL3, a component of the methyltransferase complex, and show that three of them (WTAP, METTL14, and KIAA1429) are required for methylation. Monitoring m6A levels upon WTAP depletion allowed the definition of accurate a...
متن کاملReduced m6A mRNA methylation is correlated with the progression of human cervical cancer
The m6A mRNA methylation involves in mRNA splicing, degradation and translation. Recent studies have revealed that reduced m6A mRNA methylation might promote cancer development. However, the role of m6A mRNA methylation in cervical cancer development remains unknown. Therefore, we investigated the role of m6A methylation in cervical cancer in the current study. We first evaluated the m6A mRNA m...
متن کامل5′ UTR m6A Promotes Cap-Independent Translation
Protein translation typically begins with the recruitment of the 43S ribosomal complex to the 5' cap of mRNAs by a cap-binding complex. However, some transcripts are translated in a cap-independent manner through poorly understood mechanisms. Here, we show that mRNAs containing N(6)-methyladenosine (m(6)A) in their 5' UTR can be translated in a cap-independent manner. A single 5' UTR m(6)A dire...
